If it is too short, it may not be completely cleaned, while if it is too long, it may harm the innocent. It is indeed necessary to find a precise balance point. The washing effect is related to time in a 'first rising and then stabilizing' manner, and the key is to grasp the 'effective soaking window'.

⏳ Dual phase mechanism of washing time

Rapid decontamination stage (0-30 seconds)

After adding the washing solution into the well, unbound enzyme-linked antibodies, free antigens, etc. quickly dissolve within 30 seconds (following the law of diffusion), and the OD value significantly decreases with time (the decrease can reach 60% -70%). This stage is crucial for removing most of the residual substances, as insufficient time can lead to excessively high 'initial residue'.

Slow equilibrium phase (30 seconds -2 minutes)

After 30 seconds, the concentration of residual substances in the hole has decreased to a lower level. Continuing to extend the soaking time (such as 1-2 minutes), the decrease in OD value is only 5% -10% (approaching the plateau period). At this point, the main focus is on removing 'stubborn residues' (such as low affinity non-specific complexes), but excessive extension (>2 minutes) may lead to dissociation of antigen antibody complexes that bind specifically (especially antibodies with affinity constants<10 ⁸ M ⁻¹).

🎯 Time optimization strategies for different experimental scenarios

Routine testing (standard/pure sample)

Recommended 30-60 seconds: Balancing efficiency and effectiveness, suitable for high-throughput experiments (such as washing 96 well plates within 2 hours).ELISA kitIf using a washing machine, a cycle program of '30 seconds soaking+2 seconds drying' can be set.

High background samples (such as whole blood and sputum specimens)

Extend to 60-90 seconds: Reduce non-specific background by increasing the dissolution time of 'stubborn residues'. It is recommended to use the two-step method of 'soaking, drying, and re soaking' (such as soaking for 60 seconds, drying, and immediately soaking with detergent for 30 seconds).

If the liquid in the hole dries up during the washing process (especially during manual operation), unbound substances will irreversibly adsorb to the bottom of the hole, and even if the soaking time is extended, it cannot be effectively removed (the OD value may increase by 2-3 times). Therefore, it is necessary to ensure a continuous process of 'adding liquid soaking drying', with a pause of no more than 5 seconds in between.

Collaboration with washing frequency

When the number of washes decreases (such as from 6 to 4), compensation can be made by extending the soaking time by 15-30 seconds per wash (such as the original 30 seconds and 45 seconds) to maintain the overall cleaning effect; On the contrary, when the number of washes increases, the time can be appropriately shortened (to avoid excessive dissociation).

Parameter calibration of automated washing machine

Regularly check whether the 'liquid addition soaking drying' timing of the washing machine is accurate (such as setting a 30 second soaking time, whether there is actually premature drying), and if the error exceeds ± 5 seconds, contact the engineer for calibration.
