Routine cell culture and passage process (please strictly follow aseptic operation)
1. Suck out the culture medium from the original culture bottle, rinse the cells twice with PBS buffer, and add 2-3 ml of 0.25% trypsin for digestion of the cells (pay attention to the digestion time, usually controlled within 1-2 minutes)
2. Observe the digestion under the microscope. When the cell edge shrinks and the adhesion becomes loose (it is not recommended to digest until the cell floats), remove the pancreatic enzyme, add 6-8ml of culture medium, gently blow the cell layer, and try to blow it off and disperse as much as possible.
3. Transfer a portion of the cell suspension to a new culture dish/bottle, add appropriate culture medium, mix the cell suspension evenly, and culture it in the incubator.
4. Pay attention to changes in the pH value of the culture medium, change the medium regularly (2-3 times a week), and repeat one operation or freeze storage after the cell density reaches 80%.
