1) Add the culture medium powder to 2/3 of the volume of the culture medium in double distilled water, rinse the packaging bag 2-3 times with double distilled water, stir until the powder is completely dissolved, and add the medicine according to the instructions.
HBE cell culture conditions: 90% DMEM, 10% FBS adherent cells
2) Add 0.5ml of prepared penicillin streptomycin su solution to the culture medium using a syringe, so that the final concentration of penicillin streptomycin su is 100 units/ml.
HBMEC cell culture conditions: 90% DMEM, 10% FBS adherent cells
3) Adjust the pH to around 7.2 using carbon dioxide or bicarbonate. Dilute to 1000ml and shake well.

Pulmonary neutropenia is a unified pathological manifestation in diseases such as respiratory distress syndrome (ARDS), cystic fibrosis (CF), and chronic obstructive pulmonary disease (COPD). It is speculated that the aggregation and activation of neutrophils in the lungs lead to symptoms of pulmonary dysfunction, including increased airway response, pulmonary fibrosis, and excessive mucus secretion.
Lipopolysaccharide (LPS) is the main component of the cell wall of Gram negative bacteria. When entering animal lung tissue, LPS exhibits various functions of lung damage, including white blood cell accumulation in lung tissue, pulmonary edema, severe lung inflammation, and high mortality rate.
The characteristics of LPS induced acute lung injury rat and mouse models are the accumulation of pulmonary neutrophils and the production of inflammatory mediators. Therefore, this model is very helpful for studying the targeted mechanisms related to neutrophil aggregation and mediator release.
Intranasal administration of LPS can induce an increase in neutrophils in the lungs of mice with LPS induced lung injury model. The results showed pathological changes, such as significant neutrophilic granulocytes, increased cell count, and increased production of inflammatory mediators (such as TNF - α) in bronchoalveolar lavage (BAL) fluid.
