MOC1 cell line

Thawing cell lines

1. Before thawing, add 21ml of IMDM MOC line culture medium to T150 (if you want to thaw to T75, add 10ml to T75)2. Remove liquid nitrogen from the freezer and spray a small bottle containing 70% alcohol for cleaning.

3. Place the lower part of the freezer in a water bath at 37 degrees Celsius (keep the lid out of contact with water to avoid contamination) until a small piece of ice floats.

4. Spray the cryovial with ETOH again and place it on the engine hood. Add 1ml of culture medium pipette into 1ml of cells, and add these 2ml into T150 already containing culture medium (making a total of 22ml for one T150).

5. Take some culture medium from the T150 flask, rinse the cryovial, and then place it on a plate to ensure that all residual cells remain in the cryovial.

Frozen cell line
1. Collect cells from T150

2. Rotate into particles in a 15ml conical tube (1000 RPM x 5 minutes)

3. Discharge the supernatant

4. Click on the 15ml conical tube and resuspend the cells

5. Add 1.5ml IMDM MOC culture medium and rebuild cells in the medium - maintain on ice

6. Slowly add 1.5 ml of freezing medium (prepare freezing medium -20% DMSO in IMDM MOC line medium) when pouring ice into the tube. Example: 20ml inventory - add 16ml IMDM MOC line culture medium and 4ml DMSO. Syringe filter, using um filter

7. Each serving contains 1 milliliter to 3 cryovials

8. Store at -80 degrees Celsius for no more than 2 weeks

9. Place in liquid nitrogen solution within 1-2 weeks

Note:If necessary, it can be increased to 2ml IMDM and 2ml frozen culture medium for storage in 4 small bottles. In addition, it is recommended to count cells before freezing and record them on a small bottle.
