The principle of ELISA technology is based on immunological reactions, combining the specific reactions of antigens and antibodies with the efficient catalytic effect of enzymes on substrates.
1. Solid phase immobilization of antigens or antibodies and enzyme labeling of antigens or antibodies.
2. The antigen or antibody bound to the surface of the solid-phase carrier still maintains its immunological activity.
3. Enzyme labeled antigens or antibodies retain both their immunological activity and enzyme activity.
4. The tested specimen reacts with the antigen or antibody on the surface of the solid-phase carrier, and then enzyme labeled antigen or antibody is added, which also binds to the solid-phase carrier through the reaction.
At this point, the amount of enzyme on the solid phase is proportional to the amount of the test substance in the specimen.
After adding the substrate for enzyme reaction, the substrate is catalyzed by the enzyme to become a colored product, and the amount of the product is directly related to the amount of the analyte in the sample. Therefore, qualitative or quantitative analysis can be carried out based on the depth of color. The determination method has high sensitivity (pg ng/ml level) and good reproducibility.
Experimental rules:
1. To ensure the accuracy of the pipette, the error should not exceed 2%. It can be determined using water and an electronic balance, but professional personnel will correct it.
2. Equipped with one 20ul, 50ul, 100ul, 1000ul, and one discharge gun, the gun head needs to be replaced after sucking different liquids, even when sucking standard samples.
3. One hour before the experiment, remove the reagent kit from the refrigerator and allow all reagents to return to room temperature to ensure more stable results.
4. During the experiment, the substrate should be stored away from light.
5. When using a gun to suck liquid, the speed should not be too fast to avoid the generation of bubbles and inaccurate suction volume.
6. When sucking liquid, use a gun with a range close to the required amount to reduce errors.
7. When adding liquid to the enzyme-linked immunosorbent assay (ELISA) well, avoid contact between the pipette tip and the liquid inside the well. This will allow the droplets on the pipette tip to come into contact with the well wall, and the droplets will naturally flow down.
8. After adding all the liquid, the enzyme-linked immunosorbent assay (ELISA) plate can be gently shaken parallel to the table for 30 seconds to mix the liquid. Alternatively, the shaking function of the ELISA reader can be used.
9. When taking a warm bath, use adhesive or tape to seal the enzyme-linked immunosorbent assay (ELISA) plate to prevent water evaporation.
10. When washing the plate, each time the washing solution is added, it should be left to stand for 1 minute to make the cleaning more efficient. When there is no washing machine, after pouring out the liquid, the enzyme-linked immunosorbent assay plate should be vigorously patted dry on newspaper or parchment paper.
